NMN Study: NMN is safe and increases NAD+ levels in the body

Recent research (April 2022) demonstrates that the oral administration of nicotinamide mononucleotide is safe and efficiently increases the blood levels of nicotinamide adenine dinucleotide in healthy subjects.

Summary of results:

Healthy individuals who took 125 mg of NMN twice daily for 12 weeks experienced an almost twofold increase in their whole-blood nicotinamide adenine dinucleotide (NAD) levels. NMN supplementation did not lead to any adverse physiological effects, including changes in weight or blood pressure. NAD+ levels in whole blood reached a plateau after four weeks and remained elevated throughout the 12-week study.

Summary of the research:

Nicotinamide mononucleotide (NMN) is a precursor of NAD+ that has shown beneficial effects against aging and age-related diseases in animal models when administered orally. NMN is ultimately converted into NAD+, a crucial redox cofactor that regulates numerous metabolic enzymes. NAD+ also serves as a substrate for poly(ADP-ribose) polymerase (PARP) and sirtuins, playing a role in various biological processes such as metabolism, DNA repair, gene expression, and stress responses. Previous studies in mouse models have demonstrated that NMN supplementation can increase NAD+ levels in various organs and improve the course of age-related diseases, such as obesity, diabetes, heart failure, stroke, kidney failure, and Alzheimer's disease, via NAD+-mediated pathways. However, there is still limited evidence regarding its effects on humans. In this study, we conducted a placebo-controlled, randomized, double-blind, parallel-group trial to investigate the safety of orally administered NMN and its effectiveness in increasing NAD+ levels in thirty healthy subjects. Healthy volunteers received 250 mg of NMN daily for 12 weeks (n = 15) or a placebo (n = 15), and physiological and laboratory tests were performed during this period. Additionally, NAD+ and related metabolites in whole blood were examined. Oral NMN intake for 12 weeks did not cause deviations in physiological and laboratory tests, and no clear adverse effects were observed. NAD+ levels in whole blood were significantly increased after NMN administration. We also observed a significant increase in the levels of nicotinic acid mononucleotide (NAMN), but not in NMN. Furthermore, we found that the increase in NAD+ was strongly correlated with the pulse rate before NMN administration. These findings suggest that the oral administration of NMN may be a safe and practical strategy to increase NAD+ levels in humans.
